Abstract Background and Aims Minimal change disease (MCD) is a podocytopathy more commonly seen in children, but it also accounts for 10%–25% of adult NS. Patients with MCD manifest abundant albuminuria, which related to the damage glomerular filtration barrier. Podocytes are major components barrier play crucial role maintaining integrity Alterations podocyte actin critically affect function. Tubulin polymerization-promoting protein family member 3 (TPPP3) regulator microtubule dynamic that has bundling activity. However, mechanism TPPP3 involved apoptosis remains unknown. Method Single-cell RNA sequencing comprehensively characterized expression human glomeruli. We cultured immortalized cells (HPC) vitro, constructed HPC injury model. Immunofluorescence, western blot were used determine TPPP3, α-tubulin, acetylated tubulin, phosopho-YAP1, Podocyte was determined by Annexin V APC Apoptosis Detection Kit. Results specifically expressed podocytes, its level significantly decreased cases ADR induced injury(P<0.05). Knockdown leaded decreasing tubulin podocytes(P<0.05). Phosopho-YAP1 observed knockdown HPCs(P<0.05). increased HPCs, suggesting may be podocytes apoptosis. Conclusion regulates nuclear recruitment YAP promoted acetylation.
